What are the characteristics of pregelatinized starch? How is pregelatinized starch produced?
Pregelatinized starch is a modified starch obtained by partial or complete disruption of starch granules through chemical or mechanical methods. It is commonly used as a binder, diluent, and disintegrant in tablets. Pregelatinized starch has self-lubricating properties, and when used with other excipients, magnesium stearate should be added as a lubricant. This product is non-toxic and non-irritating, but large oral doses are harmful.

1.Pre-gelatinized starch's properties;
Acidity: Acidity = 4.5-7.0 [10% (w/v) aqueous solution]
Angle of repose: 40.7
Bulk density: 0.586 g/cm³
True density: 1.516 g/cm³
Flowability: 18%-23% (Carr compressibility index)
Stability: Stable but readily absorbs moisture, should be stored in a sealed container in a cool, dry place.

Compatibility contraindications: None
Pre-gelatinized starchSolubility: Insoluble in organic solvents, slightly soluble or soluble in cold water, depending on the degree of gelatinization. Pre-gelatinized starch is sieved into well-stirred cold water to obtain starch paste; the solubility of some pre-gelatinized starch in cold water is 10%-20%.
Safety: Pre-gelatinized starch is widely used in oral solid preparations and is non-toxic and non-irritating. However, large oral doses are harmful.
2.Pre-gelatinized starchProduction:
Food-grade pre-gelatinized starch is prepared by heating a 42% (w/w) starch slurry containing water at 62-72°C. Gelling aids and surfactants should be added to control rehydration or reduce viscosity during drying. Heating forms an aqueous paste, which is then spray-dried, drum-dried, extrusion-dehydrated, or drum-dried, and finally pulverized to the desired particle size range.
Pharmaceutical fully pre-gelatinized starch does not use additives. An un-gelatinized starch aqueous suspension is spread on a hot drum, simultaneously achieving the gelatinization and drying processes. Partially pre-gelatinized starch is prepared by mechanically extruding wet starch. By pulverizing and adjusting the water content to meet standards, semi-finished products are obtained.
